WP2: Generation of a database with toxicity data from animal-free tests (in vitro data)

In WP2 a complete set of in vitro toxicology data for the 97 selected ACuteTox reference chemicals was build up to be compared with the in vivo data from WP1. The chemicals were tested by using three different basal cytotoxicity assays and five different cell lines.

The different cytotoxicity assays showed to give similar results, confirming the results from the MEIC (Multicenter Evaluation of In Vitro Cytotoxicity) project and also, the toxicity ranking of the chemicals was the same in the different assays, except for the compounds colchicine, cycloheximide, digoxin, Ochratoxin A, Thallium sulphate and Hexachlorophene. The HepG2, HL60 and NHK assays showed a higher viability than the 3T3 and Fa32 assays. Since the NR uptake assay with 3T3 cells has been validated, this assay was selected as the one used for identification of outliers in WP3.
