WP8: Development of the testing strategy
Several meeting have been held in the consortium to discuss the composition of the testing strategy. It has been agreed that the following building blocks should be consider when the testing strategy and prediction model is created:
Box 1: Pre-existing information
Biological data, existing toxicity data such as LD50, info on relevant chemicals (read across), (Q)SAR, similarity searching corrosivity, inhalation/dermal data.
Box 2: Physico-chemical properties
A. Physico-chemical properties; Molecular weight etc., High insoluability
B. Deviation in in vitro biokinetics; High volatility, Binding to plastics, Protein binding,
C. Deviations in in vivo kinetics; Oral bioavailability, Biotransformation, Distribution.
Box 3: Simple test batteries using cell lines
Basal cytotoxicity
Box 4: Biokinetic modeling
A. Absorption
B. Biotransformation
C. Distribution
Box 5: Target organ toxicity
A. Haemapoetic toxicity
B. Immunotoxicity
C. Neurotoxicity
D. Nephrotoxicity
E. Livertoxicity
The work of WP8 has been moved to WP9 and will be a part of the pre-validation exercise. The final testing strategy and prediction model will be presented in the end of the project.
